Abstract: Aim: This study aimed to determine the effects of propofol on myocardial injuries and its influence on the Sirt1/FoxO1 pathway in rats experiencing intestinal ischemia-reperfusion (I/R) injury. Methods: Rats were assigned to Sham, intestinal I/R, and three propofol groups (I/R + P1, I/R + P2, I/R + P3), receiving 10, 15, or 20 mg/kg/h propofol at reperfusion onset. The Sham and I/R groups received saline. Myocardial injury was assessed through left ventricular systolic blood pressure (LVSP), maximum increase and decrease rates of left ventricular pressure (+dp/dtmax, -dp/dtmax), serum cardiac troponin I (cTnI), creatine kinase isoenzyme (CK-MB), lactate dehydrogenase (LDH), and myocardial tissue levels of IL-1β, IL-6, TNF-α, MDA, SOD, and CAT. Protein levels were measured by Western blot. Results: I/R increased LVEDP, serum LDH, cTnI, CK-MB, IL-1β, IL-6, TNF-α, MDA, Caspase-3, Bax, and Ac-FoxO1, while decreasing LVSP, +dP/dtmax, -dP/dtmax, SOD, CAT, and survivin, Bcl-2, Sirt1. Propofol, in a dose-dependent manner, reduced LVEDP, serum LDH, cTnI, CK-MB, IL-1β, IL-6, TNF-α, MDA, Caspase-3, Bax, and Ac-FoxO1 levels while enhancing LVSP, +dP/dtmax, -dP/dtmax, SOD, CAT, survivin, Bcl-2, and Sirt1 in myocardial tissues. Conclusion: Propofol alleviates myocardial injuries caused by intestinal I/R in rats by modulating the Sirt1/FoxO1 pathway.
