Abstract: Introduction: We evaluated the effectiveness of a traditional Indian alternative medicine, Abhraka-Bhasma (nanomaterial processed biotite), with or without per-operative electrical stimulation (ES) of the repaired nerve for alleviating neuropathic pain in a rodent sciatic nerve neurotmesis and repair model. Methods: Twenty-four Sprague–Dawley rats were randomized into four groups. The control group underwent sciatic nerve division and repair alone. In group 2, per-operative ES (1 mA at 20 Htz) of the nerve was administered for one hour following nerve repair. In group 3, post-operative oral gavage of abhraka-bhasma was given for six weeks after repair, and in group 4underwent combined use of both abhraka-bhasma and ES were combined. Elevated plus maze test results and serum cortisol levels were evaluated at the second and sixth weeks. Results: A significant difference was observed between the control and the experimental groups in the EPM test with respect to open and closed-arm entry and duration, center square duration and head dipping (p<0.001) and for the two time periods (p<0.001). Serum cortisol levels were significantly different between the control and other groups (p<0.001) at both time points. The difference was also significant between the ES and the two abhraka-bhasma groups group, with the AB+ES group showing the maximum reduction near the preoperative baseline cortisol levels at the sixth week. Conclusion: Abhraka-bhasma augmented with ES resulted in enhanced recovery from neuropathic pain following neurotmesis in a rat sciatic nerve model.
