Heterocyclic Curcumin Analogues: Advances In Synthesis, Characterization, And Therapeutic Applications

Year: 2024
Type: article
Abstract: Heterocyclic curcumin analogues have emerged as promising candidates in drug discovery due to their enhanced pharmacological properties compared to native curcumin. This review explores the synthesis and characterization of oxygen- and nitrogen-containing heterocyclic curcumin derivatives, highlighting their potential in anticancer, antimicrobial, anti-inflammatory, and neuroprotective applications. Key advancements include modifications to the 1,3-dicarbonyl moiety and spacer length, which improve bioavailability, stability, and biological activity. Advanced techniques such as FTIR, NMR, and mass spectrometry have been instrumental in elucidating structure-activity relationships (SAR), enabling the rational design of multifunctional analogs. Challenges such as scalability, bioavailability, and metabolic stability remain significant barriers to clinical translation. However, recent innovations in computational modeling, delivery platforms, and translational research hold promise for overcoming these limitations. By addressing current challenges, curcumin analogs can become effective therapeutic agents for complex diseases, including cancer, neurodegenerative disorders, and infections, demonstrating their potential as next-generation natural product-based drugs.
