Abstract: Cyclic nucleotide-gated (CNG) and hyperpolarization-activated and cyclic nucleotide-modulated (HCN) channels play central roles in signaling cascades related to vision and olfaction, as well as pacemaking in the heart and brain, respectively. Understanding how these channels react to changes in their cellular environment therefore is of broad interest. To study the regulation of these channels in molecular detail, the bacterial CNG channel SthK from Spirochaeta thermophila has emerged as an important biophysical model system due to the high similarity in sequence, structure, and function to its eukaryotic homologs.
