Assessment of oxytocin and Bacillus velezensis lipopeptide on mammalian cell lines

Year: 2025
Type: article
Abstract: This study aimed to describe the effect of Bacillus velezensis lipopeptide (BVLP), and a potential but less studied nonapeptide (love and labor hormone) oxytocin (OT) as an anti-cancer and anti-obesity activity. In this study, lipopeptide and oxytocin peptides were checked for cytotoxicity and results revealed a significant reduction in 3T3-L1 (62.4 % by OT and 90 % by BVLP), Hep2 C (58.7 % by OT and 72 % by BVLP) and L20B (74 % by OT and 74.9 % by LP) in the cytotoxicity assay. MTT assays showed that BVLP significantly reduced (∼75 %) cell growth of L20B murine cell line and human cervical carcinoma, Hep2C (72 %). Besides this, changes in cellular morphology such as shrunken cell size and blebbing of the cell membrane was also observed in the BVLP (37.5 mg/ml) treated cells. The data obtained in this study was aligned with previous studies that the lipopeptides e.g., BVLP could be potential molecules to treat cancer. OD of 3T3-L1 cells at 500 nm was decreased from 1.09 to 0.9 after the treatment with BVLP while OT treatment led to increase in the OD at 500 nm from 0.96 to 1.01. The reduction in migration ability of mammalian cells has also been shown by OT treatments on various carcinomas such as Hep2C cells. In the present study addition of OT (0.155 IU/ml) to the Hep2C (cervical carcinoma cell line) cells reduced the cell migration across the gap created with the scratch.
