Abstract: Among the top 10 deadliest cancers in people, glioma is the most common form of brain cancer. Despite implementing surgical interventions, radiation therapy, and chemotherapy, the average survival period for those afflicted with this disease is approximately nine months. The ubiquitin-proteasome system (UPS) facilitates protein degradation in eukaryotic species. Proteasomes, ubiquitin, ubiquitin-like molecules, ubiquitin hydrolases, and E3 ubiquitin ligases are some of the many parts that make up the UPS. Some studies have linked UPS failure to the pathogenesis of cancer. Protein substrate stability, activity, and localization are all controlled by ubiquitination, a multi-functional post-translational modification (PTM). Dysregulation of the complex interplay between ubiquitination and deubiquitination leads to the development of several diseases, including cancer and neurological problems. There is a correlation between decreased expression of the oncogene NEDD4, which inhibits the tumor suppressor PTEN via ubiquitination, and an increased risk of getting cancer. Conversely, the overexpression of NEDD4 resulted in a reversal of these effects. This review delves into the numerous pathways that NEDD4 is involved in regarding gliomas, a critical ubiquitin ligase in these tumors. Thus, NEDD4's potential as a pharmacological treatment for glioblastoma is cause for optimism.
