Effect of pre-transplant cytoreductive therapy on the outcomes of patients with MDS or secondary AML evolving from MDS undergoing allo-HSCT: a secondary analysis of an RCT
Year: 2025
Type: article
Abstract: Allogeneic hematopoietic stem cell transplantation (allo-HSCT) remains the only curative therapy for patients with myelodysplastic syndrome (MDS) and secondary acute myeloid leukemia evolving from MDS (sAML) [ 1 ]. Multiple factors are reported to influence the prognosis of patients with MDS and sAML undergoing allo-HSCT, including prognostic stratification, disease status, conditioning regimen and pre-transplant therapy [ 2 , 3 ]. Our randomized controlled trial (RCT) has shown that granulocyte-colony stimulating factor, decitabine, busulfan and cyclophosphamide (G-DAC-BuCy) conditioning is a better choice than busulfan and cyclophosphamide (BuCy) conditioning for patients with MDS-refractory anemia with excess blasts (RAEB) or sAML undergoing allo-HSCT [ 4 ].
