In-silico primer designing and validation of Bos taurus and Sus scrofa SET proto-oncogene for non-halal detection

Year: 2025
Type: article
Abstract: SET is one of the proto-oncogenes family. Proto-oncogenes family are expressed in cell growth regulation notably during embryogenesis, wound healing, liver regeneration, and mitotic stimulation by growth factors. These genes have been widely employed in cancer detection, nevertheless, their application in halal certification assessments is minimal. Polymer Chain Reaction (PCR) has recently been an alternative method in species authentication due to its high accuracy. Primers are one of the essential components of PCR and determine the success of the amplification process. Therefore, they must be specifically designed. This research aims to explore SET proto-oncogene as a candidate primer for non-halal detection using a bioinformatics approach. SET proto-oncogene sequences from pig (Sus scrofa) and cattle (Bos taurus) were collected from the NCBI (National Center for Biotechnology Information) database. The sequences were aligned using MEGA6 software. The alignment showed areas of SET gene sequences having conserved regions and used as primer candidates. The results of this study were PCR primer forward 5’- CAA AGA ATT TCA TCT GAA TGA G -3’ and reverse 5’- CAA ATC TTT TTA CCT CTC CTT C -3’ were able to amplify the DNA of cattle with a length of 1068 bp and 1222 bp in pig.
