Smooth muscle cell Piezo1 depletion results in impaired contractile properties in murine small bowel

Abstract: Piezo1 is a mechanosensitive cation channel expressed in the intestine's smooth muscle cells (SMCs), where its role has not been investigated. We used diverse methods to determine whether Piezo1 in murine SMCs is required to mediate myogenic contractility at rest and following stretch. Using an inducible system to deplete Piezo1 from SMCs led to impaired growth and delayed motility. The Piezo1 deficient SMCs had decreased contractile strength that was asynchronous, dysrhythmic, and less frequent compared to control samples at baseline and following stretch. Moreover, ablation of SMC Piezo1 led to thinning of the muscularis, changes in the cellular composition of the epithelial layer, and a decline in pacemaker cells within the muscularis, among other changes. Our data suggest that Piezo1 in the SMCs of the small bowel is required for modulating myogenic contractile strength and rhythmicity and maintaining cell types needed to promote motility.
