Advances in DNA replication research for precision medicine

Year: 2025
Type: article
Abstract: DNA replication is a fundamental biological process ensuring genetic continuity and cellular function. Recent advances in molecular biology have deepened our understanding of the mechanisms governing DNA replication and their implications for human health. Disruptions in replication processes are linked to various diseases, particularly cancer, where replication stress and genetic instability drive tumorigenesis. Cutting-edge research, including high-resolution structural studies, single-molecule technologies, and genomic and proteomic approaches, has provided valuable insights into the molecular machinery involved in replication. These discoveries have paved the way for precision medicine applications, including targeted cancer therapies, gene editing advancements, and biomarker identification. The development of ATR and CHK1 inhibitors, as well as PARP inhibitors for BRCA-mutated cancers, exemplifies the clinical potential of DNA replication research. Despite significant progress, challenges remain in translating these findings into effective therapies due to the complexity of replication dynamics and the need for specificity in targeting replication proteins. Future research integrating multidisciplinary approaches, artificial intelligence, and computational modeling will enhance our ability to harness DNA replication knowledge for therapeutic innovation. This review highlights the critical role of DNA replication studies in advancing precision medicine and underscores the necessity of continued exploration to improve patient outcomes.
