Abstract: clinicopathological (CP) model based solely on Breslow thickness and ulceration status (AUC 0.821) (Figure 2). Figure 2. Weighted AUC (left) and C-index (right) of CP, GEP, and CP-GEP models, estimated using.632+ internal validation.Lasso and ridge regularization were applied for GEP and CP-GEP models.Input features: CP (Breslow thickness, ulceration), GEP (558 genes from discovery cohort), CP-GEP (Breslow thickness, ulceration, 558 genes). The CP model achieved higher accuracy than four other published GEP signatures. Conclusions:The study demonstrates that GEP holds significant prognostic value. To enhance its clinical utility, it is essential to focus on integrating GEP with traditional staging and CP models, enabling the development of more refined and prognostic tools.
