The Interaction between the Bone Marrow Microenvironment and Immunotherapy in Multiple Myeloma: Mechanisms, Challenges, and Prospects
Year: 2025
Type: article
Multiple myeloma (MM) remains an incurable disease. In recent years, the clinical application of immune therapies, particularly CD38 monoclonal antibodies, has significantly extended the survival of MM patients. Additionally, the emergence of innovative immunotherapies, such as chimeric antigen receptor (CAR-T) cells and bispecific antibodies (BsAbs), is reshaping the treatment paradigm of MM. However, significant challenges persist in the application of immunotherapy for MM, including tumor heterogeneity, the immunosuppressive bone marrow microenvironment, and metabolic dysregulation. Moving forward, optimizing treatment strategies that integrate targeting of the bone marrow microenvironment with immune modulation may further enhance the efficacy and safety of immunotherapies, ultimately improving outcomes for MM patients.
