Abstract: Paris polyphylla var. chinensis Smith is a medicinal herb with prominent anticancer effect by various molecular mechanisms. This study was conducted to evaluate the safety and antitumor effect of quantified dried extract of Paris polyphylla var. chinensis Smith rhizomes (PPCE) in 4T1 cell-induced breast tumor mouse model. The safety of PPCE was evaluated by acute oral toxicity test. The antitumor effect of PPCE was evaluated in BALB/c mice that were tumorigenic by injecting 4T1 breast cancer cells into the mammary adipose tissue. These mice were administered PPCE at oral doses of 100, 200, and 400 mg/kg/day or doxorubicin at an injection dose of 5 g/kg/day (once every three days) for 28 consecutive days. The study results showed that PPCE had acute oral toxicity with an LD50 of 9.73 g/kg. PPCE at a dose of 400 mg/kg significantly reduced tumor volume after 21 and 28 days of treatment with a tumor inhibition percentage of over 50% compared to the pathological group. PPCE at doses of 200 and 400 mg/kg remarkably reduced tumor weight after 28 days of treatment. However, the effects of PPCE at the tested doses on tumor histopathology were unclear. The results of this study provided evidence of the anti-breast tumor effect of PPCE in the 4T1 cell-induced breast tumor BALB/c mouse model, indicating the potential for research into the application of this medicinal plant as a product to support breast cancer treatment.
