Abstract: Background 5-Fluorouracil (5-FU) is a common antineoplastic drug used as first line of treatment in several cancers. Platelets rich plasma (PRP) is a promising therapy for soft tissue regeneration. This work aims to study the effects of PRP on 5-FU induced degeneration of the testis of adult male albino rats. Aim Evaluation of the possible effects of PRP on 5-FU induced degeneration on the testis of adult male albino rats. Materials and methods A total of 50 adult male albino rats were divided into three groups: group I (blood donor group) (10 rats): their blood was collected for PRP preparation, group II (control group) (10 rats): were held without any medication and group III (experimental group) (30 rats): allocated into three equal subgroups: subgroup III-a (5-FU group) were administrated 5-FU at a dose of 20 mg/kg daily by oral gavage for 14 days; subgroup III-b (PRP group) were administrated three weekly injections of 0.1 ml of PRP under the skin of the scrotum; and subgroup III-c (5-FU and PRP group) received 5-FU and PRP. At the expected time of each group testes were collected and processed for histological and immunohistochemical studies then morphometric study and statistical analysis were applied on the slides Results 5-FU group showed significant degeneration and reduction in the epithelial height, tubular diameter, and mean area percentage of B-cell lymphoma-2 and proliferating cell nuclear antigen stains. PRP group showed nearly normal histology. 5-FU and PRP group showed restoration of nearly normal testicular histology with significant increase in mean area percentage of proliferating cell nuclear antigen and B-cell lymphoma-2 stains as well as the epithelial height, and seminiferous tubular diameter compared 5-FU group. Conclusion PRP injection has a regenerative effect on the testicular degeneration induced by 5-FU.
