Abstract: This study aimed to assess the impact of adding glycerol to a skim milk-egg yolk extender containing butterfly pea flower extract on the quality of aceh cattle sperm during preservation. The research was conducted on aceh cattle at Regional Technical Implementation Unit for Artificial Insemination and Incubator (UPTD-IBI) Saree, Aceh Besar. A completely randomized design (CRD) was used with five treatments: glycerol was added at concentrations of 0% (P1), 4% (P2), 5% (P3), 6% (P4), and 7% (P5). The parameters examined included spermatozoa progressive motility, viability, and abnormalities at various stages: after dilution, equilibration, pre-freezing, and thawing. Data were analyzed using analysis of variance (ANOVA), followed by Duncan’s multiple range test for significant differences. Results indicated that adding different concentrations of glycerol to the extender significantly affected (P<0.05) sperm progressive motility after equilibration and pre-freezing, but not post-thawing. Glycerol also had a significant impact (P<0.05) on sperm viability after dilution and equilibration, though it did not affect viability during pre-freezing and post-thawing. The study concluded that although glycerol could not prevent the decline in sperm quality after freezing, it was effective in maintaining sperm motility during the cooling process before freezing. The best sperm motility in the pre-freezing stage was observed with a 4% glycerol concentration in the extender.
