Abstract: The development of a sensitive and rapid approach is crucial for the accurate determination of cancer markers in treatment. The present research aimed to design an electrochemical immunosensor with a sandwich-type structure to achieve highly accurate determination of carcinoembryonic antigen (CEA). The CEA has attracted the most attention in research as a protein biomarker for monitoring patients for timely diagnosis of colon cancer or the stages of disease recurrence in people who have already been treated for colorectal cancer. The MWCNT/Co(OH) 2 nanocomposite served as a modifier to improve the electrical conductivity and active surface area of the glassy carbon electrode (GCE). The Co(OH) 2 as an excellent intermediary facilitated the electron transfer on the GCE surface. This innovative immunosensor results in high sensitivity, permitting the prompt identification of CEA biomarkers in samples. Under optimal circumstances, the measurement range for CEA was broad, from 0.1 pg/mL to 100 ng/mL, while the limit of detection (LOD) was 59 fg/mL. The immunosensor demonstrated satisfactory stability, outstanding selectivity, and excellent reproducibility. • MWCNT/Co(OH) 2 nanocomposite was synthesized as a functional material. • An electrochemical immunosensor was developed based on MWCNT/Co(OH) 2 nanocomposite. • The designed immunosensor showed excellent response for Carcinoembryonic antigen (CEA) measurement. • Carcinoembryonic antigen (CEA) was detected in real serum samples.
