Abstract: Scientific advancement rests upon negations of unfounded belief systems and mooting of novel explanatory proposals in lieu. Building upon our earlier criticisms of the classical membrane pump theory (CMPT), we present irrefutable challenge to the acclaimed GHK (Goldman-Hodgkin-Katz) relation, which is based on the membrane-permeability of ≥10-1 M Na+/K+ ions and which supposedly accounts for the resting trans-membrane potential (TMP) observed in diverse cells. Thereafter, we present a simple equation of murburn-based explanation for cellular/organellar TMP, which rests on small amounts of anionic diffusible reactive species (ADRS) generated inside at ≤10-6 M. Such ADRS are spontaneously fromed from oxygen, aided by murzymes and the reducing microenvironment. The murburn (oxygen-centric) view of electrophysiology explains salient observations and hitherto inexplicable conundrums in the field, and is thermodynamically, kinetically and mechanistically validated.
