Redosing of AAV vectors in the brain after intracranial injections: Low-ish hanging fruit?
Year: 2025
Type: article
Abstract: Biologics often induce anti-drug antibodies that reduce therapeutic efficacy,1 and adeno-associated virus (AAV) vectors are no exception. Pre-existing anti-AAV antibodies are induced by exposure to naturally occurring AAV serotypes during a person’s life, and they can block the initial dosing of AAV vectors. These antibody titers vary greatly between individuals and by geography, but they tend to be of relatively low concentration. Despite this, even low titers (e.g., >1:5) have been shown to neutralize AAV vectors in preclinical models of gene therapy.
