Type: article
Abstract: Burkholderia gladioli pv. cocovenenans, an emerging foodborne pathogen, is commonly found in foodstuffs such as mushrooms and rice noodles. It produces bongkrekic acid, a highly lethal and thermally stable toxin, posing a significant threat to food safety. Therefore, there is an urgent need for the development of rapid, low-cost, and highly sensitive detection techniques. We used Si-OH magnetic beads to extract nucleic acids and loop-mediated isothermal amplification (LAMP) to amplify target DNA, effectively distinguishes between Burkholderia gladioli and Burkholderia gladioli pv. cocovenenans. The products were detected via fluorescent signals and visualized through colorimetric analysis with color analyzer software, enabling intelligent detection through a smartphone APP. The results were categorized based on color and Hue saturation lightness (HSL) values: orange (HSL ≤ 44.1) indicated a negative result, while green (HSL ≥ 63.8) signified a positive result, and the process takes less than 50 minutes, with a sensitivity of 2.5 pg/µL for pathogen-specific DNA and 6.68×102CFU/25g for detecting BGC in spiked rice noodles, 10 times more sensitive than the polymerase chain reaction (PCR), which provides technical support for the rapid detection of Burkholderia gladioli pv. cocovenenans.
