Abstract: TM treatment decreases chromatin occupancy of H3K4me3 throughout the genome and in pathways that drive ATRT oncogenesis. A. H3K4me3 shows genome wide decreases in peak height, while H3K27me3 and H3K27ac show minimal differences due to TM treatment (right panels); IgG immunoprecipitation has very small peak heights while sample immunoprecipitations have distribution of larger peak heights (left panels); B. GSEA utilizing immunoprecipitation results to calculate gene ranking shows depletion in gene sets for MYC targets, MTORC1 signaling and radial glia (stem cell) gene expression at H3K4me3 sites and depletion in reactive oxygen species at H3K27ac sites. Both changes suggest decreased expression of the noted gene sets following TM treatment. H3K27me3 gene sets did not show any significant depletion or enrichment at the FDR < 0.15 level.
