Abstract: Continued use of corticosteroids during anti–PD-L1 therapy correlates with decreased survival in a preclinical model. A, Experimental set-up. Mice bearing MC38 tumors were treated with anti–PD-L1 or isotype antibody. Mice were administered with dexamethasone or PBS intraperitoneally before (day −12 to 0) and/or during (day 0–14) anti–PD-L1 therapy. B, Tumor growth curves (mean) and survival curves in MC38 tumor-bearing mice in different treatment groups as indicated. n = 5 to 6 mice. **, P < 0.01, P values were determined by a log-rank (Mantel–Cox) test. C, Frequency of the CX3CR1+ subset in PB CD8+ T cells. PB was collected on day 14. n = 5 to 6 mice. P values were determined by the Kruskal–Wallis test with Dunn multiple comparisons. Data are presented as mean ± SEM. Ab, antibody; dex, dexamethasone; i.p., intraperitoneal; NS, not significant; αPD-L1, anti–PD-L1.
