Abstract: This study aims to determine the biological variation (difference) inherent in resting metabolic parameters in mice of different strains, so that the statistical utility of these parameters can be established for assessing physiological changes during preclinical trials and identifying significant responses to various research interventions. The aim was to study metabolic parameters in the dynamics of daylight hours in mice of the following strains: C57BL/6, CBA, Balb/c, Balb/c nude. During the daylight hours (12 hours), the metabolic intensity of mice of the following strains was studied using the MM-100 Metabolic Monitoring System and the MMCom software (USA) by indirect calorimetry. The change in average oxygen consumption (VO2, ml/h), average volume of produced carbon dioxide (VCO2, ml/h) in metabolic chambers was estimated, and the metabolic coefficient — VCO2/VO2 units was determined. These measurements were adjusted to the animal’s body weight: VO2/M and VCO2/M (M = body weight in kg). The calorimetric indicator, the intensity of heat production (IT, kcal/kg/h), was estimated taking into account oxygen consumption, carbon dioxide release per unit of time, the value of the metabolic coefficient and animal weight. Statistical processing was carried out using the Statistica 10 program. Taken together, our study provides the first systematic characterization of the basal metabolic rate in mice of the C57BL/6, CBA, Balb/c, and Balb/c nude strains, showing that mice of different strains display both similar characteristics of basal metabolism and have distinctive features not related to the influence of physical activity or the amount of calories consumed. Mice of the C57BL/6 line of both sexes have the maximum values of O2 consumption, CO2 production, and IT. CBA mice are characterized by a lower IT value, while females consume less O2 and males have higher IT than females. Female mice of the Balb/c and Balb/c nude lines have no metabolic differences, and in terms of IT value occupy an intermediate position between female mice of C57BL/6 line and the CBA line. Males of the Balb/c line are characterized by the lowest values of O2 consumption, CO2 production, and IT. The Balb/c nude line males are as close as possible to males of the CBA line according to studied indications, but, unlike them, consume more O2. Conclusions. The obtained results provide a basis for future studies aimed at understanding the mechanisms underlying metabolic variability and can be useful to pharmacologists in conducting preclinical studies of new antitumor substances.
