Abstract: This protocol describes the enzymatic assay used to identify compounds that inhibit enzymatic activity of the dengue virus protease (DENV-2 NS2B-NS3 protease). Enzymatic assay is based on the conversion of substrate into product, which is used for quantification of enzyme activity. In case of using fluorogenic substrates, cleavage by the enzyme releases a fluorescent fragment, enabling the direct quantification of enzyme activity via fluorescence detection. This assay is run as single point and dose response.
