Solubility Enhancement Techniques for Poorly Water -soluble Drugs

Year: 2025
Type: article
Abstract: It is frequently acknowledged that one of the main obstacles to contemporary pharmaceutical research is poor water solubility.A sizable fraction of recently created medication candidates is classified as Class II or IV in the Biopharmaceutics Classification System (BCS), where low solubility restricts dissolution and, as a result, therapeutic efficacy.Oral bioavailability is decreased and erratic when drugs with poor solubility dissolve slowly in the gastrointestinal fluid.Because of its high patient compliance, affordability, and ease of use, oral administration is still the recommended method for systemic drug delivery in spite of these drawbacks. On the other hand, delayed commencement of action and variable absorption patterns are frequent issues with weakly water-soluble substances.Since the pace at which these medications dissolve in the gastrointestinal tract determines their bioavailability, improving solubility is a crucial goal in formulation development. One of the best methods for enhancing dissolving behaviour among the different ways investigated is solid dispersion technology. In order to improve wettability, decrease particle size, and change crystallinity-all of which speed up dissolution-a medication is dispersed in an appropriate carrier matrix the creation, characterisation, and assessment of solid dispersions as a means of improving the solubility of medications that are not very soluble in water is the main emphasis of this study. It also covers alternate methods such complexation, pH modification, particle size reduction, surfactant use, and lipid-based formulations, providing a thorough summary of approaches to overcoming solubility-related difficulties in medication development.
