Abstract: Background: Gastric motility disorder (GMD) is characterized by delayed gastric emptying and impaired motility, significantly impacting patients' quality of life. XuanFuDaiZhe Tang (XFDZT), a traditional Chinese medicine formula, has demonstrated therapeutic effects against gastrointestinal dysfunction, but its underlying mechanisms remain unclear. Methods: Active compounds of XFDZT were identified using the TCMSP database based on oral bioavailability (OB ≥ 30%) and drug-likeness (DL ≥ 0.18). Potential therapeutic targets for GMD were retrieved from GeneCards, OMIM, TTD, DISGENET, and DrugBank databases. Common targets between XFDZT and GMD were obtained and visualized via Venn diagrams. Protein-protein interaction (PPI) networks were constructed using STRING and Cytoscape software. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were performed to explore the biological processes and pathways involved. Results: A total of 19 active ingredients and 429 common targets were identified. PPI network analysis highlighted several key hub genes. GO analysis showed enrichment in processes related to gene expression regulation, hypoxic response, and smooth muscle proliferation. KEGG pathway analysis revealed significant enrichment in the PI3K-Akt, HIF-1, and FoxO signaling pathways, all implicated in gastric smooth muscle contraction, inflammation, and neuronal regulation. Conclusion: XFDZT may exert therapeutic effects on gastric motility disorder through multi-target and multi-pathway mechanisms, particularly by modulating inflammation, smooth muscle function, and energy metabolism. This study provides a theoretical basis for the clinical application of XFDZT in treating gastric motility disorders.
