Abstract: This is a step-by-step guide to electroporation of Aiptasia (Exaiptasia diaphana) zygotes with different genetic constructs (plasmid DNA, mRNA, shRNA). It allows transformation of hundreds to thousands of zygotes in one session for anyone, who can hold and operate a micropipette. Larvae develop normally and can be used for experimentation, including establishment of symbiosis with compatible algal strains.
