Type: article
Abstract: Brown rot caused by Monilinia fructicola severely affects Shengzhou nane (Prunus salicina var. taoxingli). This study showed that treatment with 20.0 mg L−1 Natamycin (NAT) resulted in mycelial and spore deformation, including twisting and breakage, 25.0 mg L−1 NAT completely inhibited mycelial growth and spore germination. NAT exposure suppressed antioxidative enzyme activity and downregulated related gene expression, leading to the intracellular accumulation of ROS and oxidative stress. Furthermore, NAT disrupted cellular membrane integrity, causing intracellular content leakage. Mitochondrial structural damage impaired function, as indicated by decreased adenosine triphosphate content, lower energy charge levels, and reduced activities of malate dehydrogenase, succinate dehydrogenase, Ca2+/Mg2+-ATPase, and Na+/K+-ATPase. In vivo assays confirmed that NAT treatment reduced disease incidence in fruit. Additionally, NAT enhanced the activities of antioxidative enzymes, including superoxide dismutase, catalase, and peroxidase a significant reduction in MDA levels. NAT also promoted the activity of phenylalanine ammonia-lyase, chitinase, β-1, 3-glucanase, and polyphenol oxidase. The alleviation of oxidative stress and enhancement of defense enzyme activity together increased fruit resistance to M. fructicola. Consequently, higher fruit quality was achieved during storage. These findings highlight the potential of NAT treatment as an effective strategy for controlling brown rot caused by M. fructicola in Shengzhou nane.
