Design of Magnetic Mesoporous Bioactive Glass Nanoparticles for Bone Cancer Therapy: Toward Effective Hyperthermia Treatment and Bone Regeneration
Year: 2025
Type: article
Abstract: The treatment of bone cancer, particularly osteosarcoma, necessitates effective therapeutic strategies that combine tumor ablation and bone regeneration. Magnetic hyperthermia has emerged as a promising approach to selectively target malignant cells; however, developing biomaterials with optimized bioactive and magnetic properties remains a challenge. For this purpose, mesoporous magnetic and bioactive nanoparticle (MMBNs) are designed, based on superparamagnetic γ‐Fe 2 O 3 cores encapsulated in mesoporous SiO 2 –CaO bioactive glass shell. These nanoparticles are obtained through a simple multistep method involving the coprecipitation of 14 ± 3 nm maghemite particles followed by the growth of a bioactive shell via the sol–gel route. MMBNs present a uniform spheroidal morphology (180 ± 26 nm) with high specific surface area (626 ± 15 m 2 g −1 ) and excellent bioactivity, as evidenced by rapid hydroxyapatite crystallization within 3 h in simulated body fluid. This material presents superparamagnetic behavior and effective heating capacity under alternating magnetic field with clinically relevant conditions (+9 °C in 3 min), demonstrating sufficient thermal response to induce cytotoxic effects in preosteoblastic cells. The combination of rapid bioactivity and effective magnetic heating in these MMBNs suggests their high potential for integrated therapeutic strategies in bone cancer treatment.
