Abstract: CRISPR-Cas-based genome editing technology has ushered in a new era. However, Cas9, the most commonly used genome editing tool, is limited in delivery due to its large size. Type II, type V, and type VI CRISPR-Cas systems, which belong to the second largest class with Cas9, have a large number of members. Many of these effector proteins are compact and have gene editing ability that is not inferior to that of large Cas effector proteins. This review reviews the functions and mechanisms of the second major class of compact Cas effectors (type II, type V, and type VI, defined here as less than 1000 aa), summarizes their application development progress, and analyzes the problems facing the application of these compact Cas effectors in genome editing. In addition, the future development of more compact genome editing tools is discussed.
