Abstract: Background: Pain is one of the most common and immediate effects associated with insect bites (keet visha). It serves as an essential protective mechanism that signals any injury or disease, but when uncontrolled, it substan-tially affects the quality of life. Modern pharmacological treatments like opioids and non-steroidal anti-inflammatory drugs (NSAIDs) are widely used, that often come with side effects and the threat of dependency. This has lightened a growing interest in natural and herbal alternatives that provide a safer and more comprehen-sive approach to healing. The present study aimed to assess the analgesic potential of the Ayurvedic formulation Pancha Shirishanama Agad by using Haffner’sail Clip model in mice. Material and Methods: Swiss Albino mice were distributed into three groups. Group 1 (control) received distilled water, Group 2 (test drug) received Pancha Shirishanama Agad at a dose of 9.86 mg/kg, and Group 3 (standard drug) received Diclofenac sodium at a dose of 10 mg/kg. Pain response was estimated by recording response times following a tail clip impulse. Sta-tistical analysis included One-Way ANOVA with Dunnett’s post hoc test. Results: Both the test drug and Diclo-fenac sodium significantly increased response time compared to the control (p = 0.0116 and p = 0.0189, respec-tively), suggesting their notable analgesic effects. These results validate the experimental model and highlight the analgesic potential of Pancha Shirishanama Agad. Discussion: The significant analgesic potential of Pancha Shirishanama Agad validates its classical use for treating various pain-related ailments. This research serves to bridge the gap between Ayurvedic knowledge and contemporary evidence-based science, for the development of safer and plant-derived analgesic formulation to pain management.
