Response to Comment on “Trapping of Lipopolysaccharide to Promote Immunotherapy against Colorectal Cancer and Attenuate Liver Metastasis”

Abstract: We greatly appreciate Dr. Szaryńska’s interest in our work, and appreciate her comment on our article published in Advanced Materials in 2018. In a previous publication, Dr. Szaryńska’s group reported that lipopolysaccharide (LPS) increased maturation and activation status of dendritic cells (DCs) in vitro, and hypothesized that LPS may be considered a profitable supplement before launching a DC-based immunotherapy. However, the results from our publication showed that blockade of toll-like receptor 4 (TLR4) pathway or trapping of LPS by gene delivery using lipid–protamine–DNA (LPD) nanoparticles alleviated immunosuppressive microenvironment in an orthotopic colorectal cancer (CRC) mouse model, and promoted immune checkpoint therapy in CRC. Therefore, Dr. Szaryńska commented suggesting the experimental conditions and chosen aspects should be considered in clinical applications of LPS-associated protocols.
