Abstract: Oudemansiella raphanipies is a medicinal and edible fungus with various bioactivities. O.raphanipies polysaccharide (ORP) has been widely concerned due to its numerous pharmacological activities. In this study, the extraction process, physicochemical properties, structural, functional, bioactivity, and oral absorption characteristics of ORP were investigated. A homogeneous polysaccharides (UAE-ORP) was isolated from O.raphanipies by ultrasonic assisted extraction (UAE) and purified with Sephadex G-75 column. The optimal extraction process parameters (ultrasonic temperature, ultrasonic time, ultrasonic power and ultrasonic time) of ORP were optimized based on the response surface methodology (RSM). The molecular weight of UAE-ORP was 568.57 kDa with pyranose sugar ring and α-glycosidic bonds. UAE-ORP exhibited high thermal stability, implying the possibility for high-temperature food processing. UAE-ORP also showed good Function characteristics. UAE-ORP possessed strong antioxidant abilities, anti-inflammatory and prebiotic activity. UAE-ORP was significantly distributed in the gastrointestinal tract. In summary, UAE-ORP can serve as a potential source of natural functional additives.
