OIP5-AS1 Positively Regulates the Proliferation of Colorectal Cancer Cells by Repressing Apoptosis

Type: article
Abstract: Background/Aim: OIP5-AS1, a long noncoding RNA, promotes the proliferation, invasion, and migration of diverse cancer cell types such as cervical, breast, lung, and head and neck cancer. In this study, we showed that OIP5-AS1 is involved in the promotion of colorectal cancer (CRC) cell proliferation via repressing apoptosis. Materials and Methods: HCT116 and DLD-1 human CRC cells were transfected with small interfering RNAs (siRNAs) to silence OIP5-AS1. The expression levels of OIP5-AS1 in these cells were determined by quantitative reverse transcription polymerase chain reaction (qRT-PCR) and cell cycle and apoptosis analysis were performed using flow cytometry. Results: siRNA silencing of OIP5-AS1 inhibited the proliferation of HCT116 and DLD-1 cells, as well as their three-dimensional proliferation. Flow cytometry analysis showed that silencing OIP5-AS1 did not affect the cell cycle but increased the number of cells undergoing apoptosis. 5-fluorouracil (5-FU), an anticancer drug, increased OIP5-AS1 expression. Finally, we demonstrated that combination treatment with 5-FU and OIP5-AS1 knockdown dramatically inhibited the proliferation of HCT116 cells. Conclusion: OIP5-AS1 promotes the proliferation of CRC cells via repressing apoptosis and its downregulation with 5-FU treatment inhibits CRC cell proliferation more effectively.
