Andrias Davidianus-Derived Conditioner Promotes Periodontal Regeneration And Dentin Desensitization

Type: article
Abstract: Aim or purpose Periodontal regeneration represents a central focus in dental research. However, among periodontal tissues, regeneration of the periodontal ligament remains particularly challenging and understudied. In this study, we employed a natural tooth replantation model—a clinically relevant extreme scenario—to investigate the therapeutic effects and mechanisms of skin secretion of Andrias davidianus (SSAD) root conditioner, a multifunctional root conditioner, in promoting periodontal regeneration and combating dentin hypersensitivity. Materials and methods Guided by our previous studies, the SSAD root conditioner was developed through citric acid functionalization of the SSAD. In vitro, assays evaluated its effects on periodontal ligament stem cells (PDLSCs) proliferation/migration/differentiation alongside dentin remineralization capacity. In vivo, the maxillary first molar of SD rats and the anterior teeth of Beagle dogs were used to establish a natural tooth replantation model. The therapeutic effect was evaluated by histological staining, immunofluorescence, X-ray film, and CBCT. Bulk RNA-seq and scRNA-seq analyses were used to reveal the regeneration process. Results Compared with the traditional root conditioner, SSAD root conditioner has stronger viscosity, antibacterial and antioxidant capacity, and also promotes dentin remineralization. Animal studies have shown it can effectively promote periodontal tissue healing, reduce tooth loss, and enhance tooth stability. The omics map revealed it could enhance positive repair and anti-inflammatory signals and facilitate cell-cell interactions. Moreover, it could promote the adhesion between PDLSCs and the matrix by upregulating the expression of the Lamb3 gene on Laminin. Conclusions SSAD root conditioner enhances tooth replantation success by promoting periodontal tissue regeneration and alleviating dentin hypersensitivity.
