Abstract: Background and Aims: Progression to acute myeloid leukemia (AML) is detected in approximately 20% of patients with congenital neutropenia (CN) after 20 years and is frequently associated with co-occurring mutations in RUNX1 and CSF3R. The hypothesis sets CN as a pre-leukemic condition, in which the acquisition of somatic mutations in RUNX1 and CSF3R drives leukemia progression. Here, we aim to establish an in vivo model of CN-neoplasms to investigate the clonal architecture induced by the accumulation of cooperative somatic mutations and their leukemogenic potential.
