Type: article
Abstract: Artocarpus lacucha , a tropical tree traditionally used in Southeast Asian medicine, has attracted scientific interest due to its bioactive components, particularly oxyresveratrol (OXY). This study evaluated the ethanolic extract of A. lacucha (AE) for its antioxidant, anti-inflammatory, neuroprotective and anti-aging properties. The AE, containing approximately 45 % OXY, exhibited potent antioxidant activity (IC 50 values of 4.55 and 2.13 µg/mL for DPPH and ABTS assays, respectively) and inhibited nitric oxide (NO) production in LPS-stimulated RAW 264.7 cells (IC 50 = 12.17 µg/mL). In SH-SY5Y neuronal cells, the AE demonstrated a protective effect against hydrogen peroxide (H₂O₂)-induced cytotoxicity (restoring viability to approximately 80 % at 6.25–25 µg/mL) and Aβ 25–35 -induced toxicity (improving viability at 6.25–12.5 µg/mL). In Caenorhabditis elegans , AE treatment at 25 and 50 µg/mL extended mean lifespan by 29.33 % and 20.64 %, respectively, compared to the control group. Additionally, AE at 25 and 50 µg/mL significantly improved resistance to heat, UV radiation, and oxidative stress. These findings highlight A. lacucha extract’s potential as a therapeutic agent for aging and Alzheimer’s disease, warranting further mechanistic and clinical studies. • AE protected SH-SY5Y neuronal cells against H 2 O 2 and Aβ₂₅₋₃₅-induced cytotoxicity. • AE extended the mean lifespan of C. elegans , highlighting its anti-aging potential. • AE significantly protected C. elegans to UV radiation and oxidative stress.
