Type: article
Abstract: Significance Discuss the effects and mechanisms of Nrf2 in skin damage due to psoriasis. Approach A psoriasis-like mouse model and HaCaT cell model were established, followed by overexpression and interference of Nrf2 and NLRP3 genes. Immunofluorescence histochemistry was used to detect the expression level of TXNIP in skin lesions. The levels of IL-1β and IL-18 in both mouse and cell groups were measured using enzyme-linked immunosorbent assay (ELISA). Quantitative real-time PCR (qRT-PCR) and Western blotting were conducted to detect the gene and protein expression levels of Caspase-1, IL-1β, IL-18, TRX1, and NLRP3. Results Intervention with si-NLRP3 or Lv-Nrf2 significantly improved PASI scores, while the expression levels of Caspase-1, TRX1, NLRP3, IL-1β, and IL-18 were markedly decreased. In vitro, Lv-Nrf2 intervention significantly improved IL-1β and IL-18 concentrations, and the expression levels of Caspase-1, TRX1, NLRP3, IL-1β, and IL-18 were significantly downregulated. Conclusions Nrf2 could reduces psoriasis skin damage by inhibiting NLRP3
