A2|sELISA for identification of biological markers 2415

Year: 2025
Type: article
Abstract: Description The identification of relevant biomarkers in biological matrices or fluids is paramount to bolster the understanding of disease pathogenesis. The A2 microwell multiplex arrays designed for sandwich enzyme-linked immunosorbent assays (A2®|sELISA) are robust and cost-effective for profiling biomarkers. The goal of the study is to apply the of A2|sELISA multiplex technology for identification of biological markers in clinical specimens. Here, we describe the utility and practicality of the multiplex, plate-based assay platform to assess biomarkers in serum and cerebrospinal fluid (CSF) samples derived from patient participants with immunological and autoimmune (eg., multiple sclerosis and amyotrophic lateral sclerosis) disorders and healthy controls. Capture antibodies against immune regulatory markers were imprinted in a multiplex array format into each well of a 96-well plate. Serum or CSF samples derived from patients and healthy controls were subjected to multiplex sELISA arrays. Data and statistical analyses of the mean pixel intensity read-outs from the multiplex A2|sELISAs were performed using a validated statistical software. Initial application of the technology suggests A2|sELISA method is an efficient and cost-saving method for profiling alterations in biomarkers present in biological matrices derived from patient versus control samples.
