Abstract: Description With cancer rates in the world on the rise novel therapeutics are needed to help combat this trend. Two major problems with new therapies are cost and safety. Live attenuated bacteria address these limitations. Previously we have shown that live attenuated bacterial therapeutics display efficacy by delaying the growth of tumors and increasing survival in experimental murine models of cancer. We have now engineered a microbe to promote increased efficacy by engineering bacterial secretion systems able to deliver therapeutic cargo outside of the cell while keeping the cargo bound to the bacterial cell surface, thereby establishing proof-of-concept for the production and secretion of bacterial-derived cargoes. In future work, we will further engineer the microbe to secrete immunologically active molecules with the aim of improving efficacy.
