Abstract: Description Growing evidence suggests that the gut microbiome significantly impacts the health, metabolism, and immune system of its host. Modulating commensal bacteria within the human gut has demonstrated therapeutic potential for treating intestinal diseases, including inflammatory bowel disease (IBD). In this study, we evaluated the therapeutic potential of Lactobacillus paracasei KBL382, a probiotic bacterium isolated from human fecal samples, in pre-clinical models of intestinal disease. Our results showed that KBL382 reduced pro-inflammatory cytokines and increased anti-inflammatory cytokines in human peripheral blood mononuclear cells (PBMCs). KBL382 significantly alleviated symptoms in both dextran sulfate sodium (DSS)- and 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced IBD mouse models. The in vivo effects of KBL382 were further validated through intestinal permeability assays and the analysis of molecular markers. Additionally, the potential benefits of KBL382 were reflected in positive changes in gut microbiome alpha and beta diversities. Collectively, these findings underscore the promising therapeutic potential of Lactobacillus paracasei KBL382 in the treatment of IBD. Topic Categories Therapeutic Approaches to Autoimmunity (THER)
