Abstract: Description Aging is associated with a decline in the immune system (termed “immunosenescence”) as well as enhanced inflammation, known as inflammaging, both of which are linked to metabolic disorders like diabetes. Metformin, a type 2 diabetes medication, is senomorphic, and has been shown to improve aging markers such as cellular senescence. We previously demonstrated that senescent peripheral CD8+ T cells show increased senescence-associated β-galactosidase activity and fail to function. The link between diabetes, aging, and senescence shows the need for integrated approaches to tackle age-related diseases. To test if CD8+ T cell senescence correlates with diabetes, blood samples from older individuals (ages 65 and up) were analyzed for immune cell parameters and diabetes indicators (HbA1c levels using A1C Now Self Check kit). Data revealed that older donors had significantly more senescent PBMC than younger donors (ages 18-30). Additionally, higher levels of CD4+ and CD8+ T cell senescence were observed in individuals with prediabetic A1C levels (5.7- 6.4%), while metformin treatment was correlated with reduced CD8+ T cell senescence. These findings indicate a significant relationship between elevated HbA1c levels and immunosenescence, supporting a link between immune aging, diabetes, and metformin treatment. Further analyses will explore additional senescent markers and diabetes indicators and treatments.? Topic Categories Lymphocyte Differentiation and Peripheral Maintenance (LYM)
