Real-time visualization of neutrophil distribution in the thymus following CXCR4 antagonist (plerixafor) treatment using intravital microscopy 3656

Abstract: Description Plerixafor, a CXCR4 antagonist, is commonly used in clinical treatments and promotes neutrophil production by acting on hematopoietic stem cells. However, current studies on its pharmacokinetics (PK) and mechanisms often rely on endpoint data from splenectomy, peripheral blood analysis, or histopathology, which are limited by their reliance on data collected at the time of animal sacrifice. This approach does not account for whether neutrophil accumulation is a direct response to treatment or influenced by pre-existing stress, leading to ambiguity in understanding immune responses. To overcome these limitations, this study utilized intravital microscopy to provide real-time imaging of neutrophil dynamics in vivo. Animals were divided into two groups, with two animals per group. Imaging was performed at two thymus locations at 0, 45, and 90 minutes after plerixafor treatment, enabling precise observation of neutrophil production and accumulation. Results showed that neutrophil accumulation in the thymus could be observed in real time, consistent with previously reported data. Intravital microscopy provided a more robust and dynamic understanding of neutrophil reconstitution and supported prior findings with greater scientific precision. This technique also offers the potential to uncover previously unexplained molecular mechanisms in immune system dynamics, making it a valuable tool for studying the effects of CXCR4 antagonists like plerixafor and advancing immune research.
