Applying cytek spectral flow cytometry for comprehensive phenotyping of Acute Myeloid Leukemia (AML) in whole blood and procured samples 2737

Abstract: Description Acute Myeloid Leukemia (AML) is an aggressive hematologic malignancy that arises from blood-forming stem cells and is characterized by substantial genetic diversity and varying levels of myeloid differentiation. Accurate phenotyping is crucial for diagnosis, prognosis, and treatment strategies. However, conventional flow cytometry is restricted by its limited multiplexing capabilities and the complexity of compensating for fluorescence spillover. Spectral flow cytometry overcomes these limitations by using a full spectrum of fluorescence to enhance the resolution and sensitivity of phenotyping studies. We used the Cytek spectral flow cytometer to perform a comprehensive immunophenotyping of AML with experiments including titrations and the selection of specific clones to optimize the detection of relevant markers of interest. The panel incorporates a comprehensive array of markers critical for distinguishing progenitor and stem cells, leukemic stem cells (LSCs), activation and co-stimulatory molecules, along with lineage-specific and backbone markers. The use of whole blood and procured AML samples ensures that our findings are robust and applicable to clinical settings. Future work will focus on validating these phenotyping results across a larger cohort of AML samples and integrating the spectral flow cytometry data with genomic and proteomic analyses to develop comprehensive diagnostic and prognostic profiles.
