Abstract: Description NKG2A is the subject of intense study in immuno-oncology for its role as a checkpoint receptor on human natural killer (NK) cells. NKG2 receptor family members bind to HLA-E and can provide either inhibitory or activating signals based on their intracellular domains. Accurate identification and expression analysis of individual NKG2 receptor types is therefore pivotal for understanding their respective biological role(s) in NK cell function. However, generating high-affinity NKG2A-specific antibody with low cross-reactivity to other family members has been challenging due to high homology in their C-type lectin ectodomains. Here, we introduce a new recombinant monoclonal antibody, clone DD11-701.rMAb, that is highly specific for human NKG2A and shows improved resolution when compared to other commercially available clones. This new antibody demonstrates excellent resolution, effectively detecting NKG2A-expressing CD56bright NK cells and CD3+ T cell subsets in a multicolor flow cytometry panel. Our results also show this antibody specifically stains NKG2A-expressing 293 cells but not those expressing NKG2C. This antibody will be available in a wide range of fluorochrome formats allowing for simpler panel design for flow analysis and cell isolation. Clone DD11-701.rMAb meets the need for a highly specific NKG2A antibody, empowering researchers to conduct basic and translational research aimed at advancing NK cell-based immunotherapy.
