Development of an ex-vivo transmission blocking assay for Plasmodium field-isolated gametocytes

Abstract: For the successful control of malaria, there is an urgent need to identify safe transmission-blocking therapies and tools that reliably measure the blocking properties of these therapies. The current tools for the assessment of transmission-blocking drug activity are technically challenging, challenging to implement in the LMIC laboratories. Besides, the existing tools often rely on conventional laboratory strains which may not be reflective of the plasmodial population circulating in malaria-endemic regions. The development of robust assay using Plasmodium isolates and implementable in most LMIC laboratories are crucial for drug discovery. Our project aims to develop an assay to measure the transmission-blocking activity drugs by exploiting parasite isolated from malaria infected-volunteers. Parasites will be exposed to selected drugs, and the transmission-blocking capability will be evaluated using the gold standard mosquito feeding assay. This innovative work is anticipated to be a useful addition to the current tools for drug discovery and to support the malaria eradication agenda.
