The Road to the Full Sequencing of Natural Frogs’ Peptides Relying Solely on Mass Spectrometry

Abstract: Amphibians, as one of the leaders of immune resistance, live on Earth for hundreds of millions of years.Their dorsal glands produce a cocktail of biologically active peptides, successfully fighting microorganisms and even predators.Since this mechanism prevents development of the pathogens' resistance, antimicrobial peptides are very perspective pharmaceuticals of future generations.Mass spectrometry is the most powerful tool to sequence peptides/proteins.For over 30 years of studies in that field mass spectrometry resolved all the problems with the de novo sequencing of amphibian peptides. This review covers the modern de novo sequencing algorithms enabling achieving complete sequence coverage of all frog peptides including long (up to 46 aa) ones.Accurate mass measurements have reliably solved the problem of isobaric amino acids. Moreover, there is no need any more to carry on any preliminary derivatization procedures like the brake of disulfide bonds or N-terminal acetylation.EThcD and ExD tools with manual spectra interpretation provide an efficient approach of reliable differentiation between isomeric Leucine/Ileucine residues in the chain, using secondary w-and d-ions, and resolve the problems of the sequence inside the intact S-S cycles.
