Abstract: ABSTRACT Polyphenols, natural compounds abundantly distributed in plants, possess potent antioxidant and anti‐inflammatory activities and hold promising potential in cardiovascular protection and the management of metabolic disorders. Nevertheless, their intrinsic instability and low bioavailability severely constrain their practical and large‐scale applications. Polysaccharide‐based nano‐delivery systems address these challenges by enhancing stability and enabling targeted release. This protocol centers on the preparation of polyphenol‐polysaccharide nanoparticles, emphasizing four core techniques‐ionic gelation, antisolvent precipitation, self‐assembly, and nano spray‐drying‐and provides an in‐depth analysis of their mechanisms, procedural steps, and key parameters. Furthermore, the protocol presents a comparative table summarizing each method's precautions, applicable materials, features, and limitations. Synthesizing current research evidence, this protocol serves as a standardized reference for nanoparticle preparation, assisting researchers in optimizing process parameters and improving product consistency, thereby fostering innovation and standardization in applications such as functional food additives, targeted drug delivery, and green agricultural carriers.
