Abstract: In this study we report on the development of an innovative cotton thread-based colorimetric sensor coupled to smartphone digital image-based (DIB) treatment for the quantification of L-ascorbic acid (L-AA). The designed microfluidic cotton thread-based analytical device (μTAD) employs a redox reaction involving Prussian Blue (PB), which reduces the blue color from PB due to the presence of L-AA. The experimental conditions were optimized using a central composite design to establish ideal conditions for sensor fabrication. The microdevice, which requires only 30 µL of sample, demonstrated a good linearity (R² = 0.993), a low limit of detection (0.18 mg L⁻¹), and satisfactory recovery rates from 87.2% to 106% for complex sample such as fruit pulps. Moreover, using iodometric titration as a reference method, no significant differences at a 95% confidence level were obtained, confirming the reliability and effectiveness of the proposed method.
