NLRP3 inflammasome inhibition attenuates liver injury and dysfunction induced by noise exposure in rats

Abstract: Noise, a significant environmental stressor, can induce dysregulated glucose and lipid metabolism. However, the precise molecular mechanisms driving liver dysfunction in noise-induced dysregulated glucose and lipid metabolism remain incompletely elucidated. Emerging evidence has highlighted the pivotal part played by NLRP3 inflammasome (Nod-like receptor protein 3) in liver pathology. We established a noise-exposure rat model (100 dB, 4 hours/day for 30 days) to determine whether NLRP3 inflammasome drove noise-caused hepatic injury and dysfunction. Noise exposure rats exhibited notable alterations in liver architecture and dysfunction. Moreover, our study showed NLRP3 inflammasome assembly activation, which was reversed after NLRP3 inhibitor MCC950 intervention, mitigating liver damage and improved liver function. In conclusion, our investigation suggested the NLRP3 inflammasome activation instigated by noise exposure, which promoted liver inflammation, thereby causing hepatic impairment and dysregulated glucose and lipid metabolism. These data present novel mechanistic viewpoints about the pathogenesis of noise-caused hepatic damage and dysfunction.
