Automated Scaffolded Peptide Synthesis Optimization via Multi-Modal Data Evaluation and HyperScore-Driven Feedback

Year: 2025
Type: article
Abstract: **Abstract:** This paper presents a novel framework, *Automated Scaffolded Peptide Synthesis Optimization (ASPSO)*, for streamlining and accelerating the discovery and optimization of biologically active peptide therapeutics. ASPSO leverages multi-modal data ingestion, advanced semantic parsing, rigorous logical consistency and verification, and a hyper-score-driven feedback loop to predict and optimize peptide synthesis conditions in silico, drastically reducing experimental costs and timelines. Current peptide synthesis methods are often iterative and resource-intensive, requiring substantial empirical validation. ASPSO aims to overcome these limitations by integrating textual data (scientific literature), structural information (peptide sequences and 3D conformations), and computational simulations (chemical kinetics & thermodynamics) into a unified evaluation pipeline. The estimated market size for peptide therapeutics is projected to reach $13.8 billion by 2028. We demonstrate the potential for a 10x reduction in experimental iterations required for optimizing peptide synthesis, leading to significant cost savings and accelerated drug discovery. --- *This document is a part of the Freederia Research Archive.
